How to establish a stable and reproducible antibody quantification method has long been a major focus in the field of neuroimmune research.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Against this background, the Uni Pharma Molecular Biology Laboratory has launched an industry-academia collaboration with the Institute of Clinical Medicine at National Yang Ming Chiao Tung University (NYCU). Together, they have established a quantitative autoantibody research platform that combines Live Cell-Based Assays with Flow Cytometry, aiming to provide a novel technical tool and data foundation for related research.<\/p>\n\n\n\n<h2 class=\"wp-block-heading has-large-font-size\"><strong>Research Background: Technical Exploration from Qualitative to Quantitative Methods<\/strong><\/h2>\n\n\n\n<p class=\"wp-block-paragraph\">Although currently common techniques such as the Immunofluorescence Test (IFT) and Cell-Based Assay (CBA) have been widely applied in research and clinical adjunctive diagnosis, their results are mostly presented as positive\/negative or as titers. This presents technical limitations in terms of numerical resolution, cross-batch consistency, and long-term monitoring applications.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">Therefore, this collaborative research focuses on establishing an analytical workflow capable of numerically representing antibody reaction intensity with excellent reproducibility, serving as a foundation for subsequent research and methodological validation.<\/p>\n\n\n\n<h2 class=\"wp-block-heading has-large-font-size\"><strong>Technological Concept: High-Transfection Live Cell Platform Combined with Flow Cytometry<\/strong><\/h2>\n\n\n\n<p class=\"wp-block-paragraph\">This research platform is based on HEK 293T cells expressing specific antigens. By optimizing transfection conditions or utilizing cell lines with stable gene expression, the antigen expression rate on the cell surface is maintained at a highly stable and high-proportion level. Flow cytometry is then used to measure the distribution of fluorescent signals after antibody-antigen binding, further establishing a consistent quantitative analysis workflow.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">In addition to optimizing cell stability across batches, the research team simultaneously developed calibration and computational methods to reduce the impact of non-specific signals in serum samples on the analysis results, thereby enhancing the overall reliability and reproducibility of the data.<\/p>\n\n\n\n<h2 class=\"wp-block-heading has-large-font-size\"><strong>Research Observations: Stability and Consistency of the Quantitative Analysis Workflow<\/strong><\/h2>\n\n\n\n<p class=\"wp-block-paragraph\">During the research process, the team analyzed different specimen types and observed the following:<\/p>\n\n\n\n<ul class=\"wp-block-list\">\n<li>Flow cytometry can generate stable and reproducible signal distribution curves.<\/li>\n\n\n\n<li>Quantitative analysis results show a consistent trend across different batches of experiments.<\/li>\n\n\n\n<li>Quantitative values can be used to reflect changes in antibody reaction intensity, serving as an observational indicator for research.<\/li>\n<\/ul>\n\n\n\n<p class=\"wp-block-paragraph\">Related results indicate that this platform demonstrates feasibility as a research-grade tool for antibody quantification. Moving forward, more research data needs to be accumulated to further validate its scope of application and analytical characteristics.<\/p>\n\n\n\n<h2 class=\"wp-block-heading has-large-font-size\"><strong>From a Single Antigen to a Multiplex Neuroimmune Research Platform<\/strong><\/h2>\n\n\n\n<p class=\"wp-block-paragraph\">Currently, the research platform has completed the construction of a quantitative analysis workflow centered on the NMDAR1 antigen, and is gradually expanding its research applications to other neuroimmune-related antigens. In the future, as research data accumulates and methodologies continue to be optimized, the platform is expected to become an important technical resource for exploring changes in antibody responses in neuroimmune research.<\/p>\n\n\n\n<p class=\"wp-block-paragraph\">The Uni Pharma Molecular Biology Laboratory and NYCU will continue to deepen their industry-academia collaboration to promote the development of autoantibody quantification technologies in the research field.
